中文摘要: 人体的衰老以及绝大多数疾病的发生都与活性氧自由基的过量产生有直接或间接的联系。清除活性氧自由基是天然抗氧化剂的最主要抗氧化机制。然而在实际的食品或细胞等复杂体系中,受环境以及其它共存抗氧化剂的影响,天然抗氧化剂的抗氧化分子机制常常更为复杂。我们按计划选择了一系列结构关联的天然类黄酮化合物为研究对象,采用稳态和时间分辨光谱学手段结合化学合成、电化学和理论计算分析,系统研究了类黄酮化合物在非均相脂质体中的分布、与膜的相互作用和分子结构(黄酮与异黄酮骨架,羟基的位置和个数)、反应动力学以及与另一类天然抗氧化剂类胡萝卜素的协同、拮抗作用等对抗氧化活性的影响和构效关系规律。基于类黄酮化合物与类胡萝卜素分子间的相互作用,设计合成了一种以类黄酮化合物和类胡萝卜素为导化合物的新型抗氧化剂。此外,借助本项目的支持,还创新性的开展了类胡萝卜与寿命最短、氧化性最强的活性氧自由基HOo的直接、快速自由基反应机理的研究。本项目所取得的研究结果可以为天然抗氧化剂的筛选、提高抗氧化效率提供理论指导。

英文摘要: Reactive oxygen radicals are found more or less, directly or directly related to the human ageing and most diseases. For natural antioxidants, radcial scavenging is considered the most prominent antioxidation mechanism. In food or cell systems, the molecular mechanism of the reactions between natural antioxidants and reactive oxygen radicals is important for the evaluation and the application of antioxidants. In present study, a series of structure-related naturally occuring flavonoids were investigated in both homogeneous solvent and heterogeneous liposome by laser flash photolysis combined with chemical synthesis, electrochemical measurements and quantum chemical calculations to elucidate the relationship between antioxidant efficiency and structures, dynamics and the interactions (synergism or antagnism) of flavononids with another kind of natural antioxidant carotenoid.A novel antioxidant retinylisoflavonoid was synthesized by using of a flavonoid and a carotenoid as precursors. In addidtion, a fast radical reaction of carotenoids with the most short-lived and oxidative reactive oxygen species HOo was also investigated.
