中文摘要: 在前期研究工作中发现,肠炎沙门氏菌基因组中的原噬菌体SE14(ΦSE14)对它在蛋清中的存活能力具有促进作用。本研究将以肠炎沙门氏菌ΦSE14以及该噬菌体中具有抗逆性的特异基因SEN1383和SEN1393为研究对象,通过构建突变株,分析ΦSE14以及特异基因对该菌在蛋清中存活的影响;通过细胞形态、胞壁结构、运动能力、群体效应等表型,分析表型对该菌在蛋清中存活的影响;通过高通量mRNA测序,结合表型差异结果,寻找影响表型差异或与抗逆性相关的关键基因,分析关键基因对该菌在蛋清中存活的影响;通过生物信息学预测和生物学验证,寻找到与特异基因、ΦSE14和关键基因相关的小RNA,分析小RNA对该菌在蛋清中存活的影响;通过分析小RNA、关键基因以及与ΦSE14相互调节作用,初步解析ΦSE14促进肠炎沙门氏菌在蛋清中存活的分子机制。

英文摘要: In previous studies, it was found that Salmonella Enteritidis has a high survival rate in egg white, and there was a prophage SE14 (ΦSE14) in its genome which could promote the survival viability of this serovar. Here we will focus on this survival molecular mechanism mediated by ΦSE14. Besides the ΦSE14 deletion mutant we constructed before, two Salmonella Enteritidis specific genes (SEN1383 and SEN1393) will be deleted in this study. These key genes synergistic with ΦSE14 and/or Salmonella Enteritidis specific genes will be determined by the combination of their phenotypic differences analysis and mRNA sequencing. Several small RNA regulated these above key genes will be evaluated by bioinformatics prediction and RNA silencing. In this way, the role of ΦSE14 and Salmonella Enteritidis specific genes played in its survival mechanism in egg white will be revealed.
