中文摘要: 胰腺癌致死率极高,早期诊断水平直接影响患者的治疗效果及生存质量,而现有的标志物特异性和时效性差,且检测技术亟待提升。我们前期已经发现胰腺癌患者的外周血中存在胰蛋白酶原基因(PRSS1)突变-可以作为易感人群筛查的分子标志,而且血清胰蛋白酶的异常增高与早期胰腺癌密切相关。胰蛋白酶可通过降解细胞外基质参与胰腺癌的侵袭和转移过程,还可以作为信号分子激活PAR-2刺激胰腺癌细胞的增殖和黏附。因此PRSS1基因突变和胰蛋白酶联合检测对胰腺癌的早期诊断具有重要意义。本项目拟利用混合探针构建多基因识别界面,结合纳米电化学传感技术组装成同一界面同时检测多个基因位点的分子开关型传感器,用于检测PRSS1等胰腺癌易感基因,并利用抗原-抗体的特异性结合原理构建双抗体夹心型电化学免疫传感器检测血清胰蛋白酶,以期在基因易感性筛查的基础上同时监测血清胰蛋白酶等指标,及时、准确地评估高危人群罹患胰腺癌的风险。

英文摘要: The high mortality rate of pancreatic cancer is a bottleneck for further treatment with long-term efficacy. Thus, it is urgent to identify new methods to accurately predict the early onset of pancreatic cancer. Our previous studies have found that trypsinogen gene (PRSS1) mutations in the peripheral blood of patients with pancreatic cancer and it can can be used as the susceptible molecular marker in population screening. And abnormally high level of serum trypsin is closely related to early pancreatic cancer. Trypsin involved in pancreatic cancer invasion and metastasis through the degradation of extracellular matrix. It also serve as signal molecule to activate PAR-2 and stimulate pancreatic cancer cell proliferation and adhesion. So, the combined detection of PRSS1 gene mutations and trypsin is significant important to diagnosis of pancreatic cancer. On the basis of these studies, we will use mixed probe to build a multi-gene recognition interface to detect PRSS1 mutations. And it will combinate with nanoelectrochemistry sensing technology to detect multiple genes on the same interface molecular switch sensor.
