中文摘要: 能源危机、资源匮乏,是21世纪急需缓解的难题。光合作用赋予我们巨大的生物质资源,如何有效利用这些生物质将其转化为生物能源,对人类可持续发展具有非常重要的意义。本项目以能产生细胞壁降解酶的菌株Q7-31为材料,研究其植物细胞壁降解酶酶系组分、关键酶及最佳比例、协同作用、酶的结构等理论方面;通过酶谱鉴定测定酶氮末端氨基酸序列、克隆出5-15种关键酶并在大肠杆菌中进行克隆与表达;研究这些关键酶的酶学性质、酶促动力学等。从基因-蛋白质水平揭示植物细胞壁降解酶的结构与功能及其基因的表达调控规律。本研究有助于揭示基因、蛋白质和细胞壁降解之间的关系,并为其他类型的降解酶的研究提供借鉴。解决目前生物能源研究中的酶降解效率底及多种酶降解中酶最佳配比不详等问题;Q7-31分离自青藏高原,为丰富青藏高原真菌资源的基础研究提供蛋白质水平的信息。为丰富青藏高原真菌资源的基础研究提供基因、蛋白质水平的信息。

英文摘要: There are many difficult problems for human being in 21 century, like energy crisis, lack of the resources, while a large amount of plant biomass source on earth is produced the photosynthesis every year. It plays very important roles for sustainable development of human being to make use of these plant biomass sources into bio-energy. In this study, the components of the enzyme from the strain of Fusarium sp. Q7-31 that could produce cell wall degrading enzyme (CWDE), their key enzymes and synergistic functions will be studied. Genes of CWDE were cloned and expressed in E.coli; the ratios of different enzyme from CWDE and the characteristics and reaction kinetic of these enzymes were studied. The expression and regulation of genes from CWDE and their structure and function would be deeply studied at the level of gene-protein. This studied will be helpful for the revealing the relationship of gene, protein and degradation of the cell wall function and also providing some models for other types of enzyme studies. It also could solve some problems of low effect and enrich the knowledges of enzyme ratios in the bio-energy research. The strain of Q7-31were isolated from Qinghai-Tibetan Platea, it also provide more information on gene and protein for the fungi resoures from Qinghai-Tibetan Platea.
