中文摘要: Ⅲ型前列腺炎是由多种炎性基因共同作用的自身免疫性疾病。miRNAs可通过调控多个信号网络通路对炎症反应和免疫反应起重要调节作用。我们前期研究发现miRNAs在Ⅲ型前列腺炎中呈现差异性表达,解毒活血汤治疗Ⅲ型前列腺炎疗效显著,能有效调节前列腺组织中细胞因子水平。 本课题以我院有效治疗Ⅲ型前列腺炎的解毒活血汤为研究对象,运用miRNA芯片技术、RT-PCR技术、Northern blot技术检测miRNAs在Ⅲ型前列腺炎大鼠前列腺组织中的表达情况,筛选出特异性miRNA,并结合生物信息学方法对特异性miRNA靶信使基因进行功能富集分析,获得miRNA靶基因和基因功能调控网络。同时研究解毒活血汤对它们表达的干预作用,探讨解毒活血汤治疗Ⅲ型前列腺炎的关键作用机制,为中医药防治前列腺炎提供理论和实验依据。

英文摘要: Type Ⅲ prostatitis is a kind of autoimmune disease which is interacted by various inflammatory genes.MiRNAs regulate the response of inflammation and immune by multiple signaling network pathways.According to our previous study found differentially expressed miRNAs presented type Ⅲ prostatitis. Jieduhuoxue Tang has certain effect in the treatment of type Ⅲ prostatitic.It can adjust the levels of cytokines'expression in prostatic tissue effectively. This study uses miRNA chip technology 、RT-PCR technology 、Northern blot to test miRNAs'expression in prostate tissue of type Ⅲ prostatitis rat model,in order to filter out specific miRNA.And conbining with bioinformatics methods to functional enrichment analysis the target messenger genes of specific miRNA,to get miRNA target genes and gene function regulatory networks.At the same time,we observe the intervention effect of Jieduhuoxue Tang on their expression ,explorating the key mechanism of Chinese medicine treatment for type Ⅲ prostatitis , providing theoretical and experimental evidence for the treament and prevention of Chinese medicine for prostatitis.
