中文摘要: 纳米金颗粒由于其优良的稳定性、特殊的光电效应以及优良的生物相容性,在药物靶向传输体系、疾病检测、分子识别、生物标定、化学催化以及光电学领域有着良好的应用前景。小粒径的金纳米簇(AuNCs)由少数几个金原子组成,它能够稳定存在,并且随着纳米簇中原子数的改变,能发射从可见光到近红外光范围的荧光。在本项目中拟设计合成出新的多巯基化合物,以硼氢化钠为还原剂,通过一锅法制备近红外发光的水溶性金纳米簇。多巯基化合物的多齿配位效应,为纳米金团簇提供了单分散性和优良的胶体稳定性。同时,在金纳米簇外面包裹磷脂构建金纳米簇脂质体,进一步增加其稳定性和生物相容性。脂质体表面修饰肿瘤靶向多肽分子,在多肽的介导作用下,实现特定肿瘤的诊断。研究成果将为采用无机纳米材料进行肿瘤诊断提供一种新的思路。

英文摘要: Gold nanoparticles have a great protential application such as target drug delivery, desease detection, molecular recognition, cell labeling, chemical catalysis and optoelectronic, due to their good stability, unique photoelectric effect and excellent biocompatibility. Small gold nanoclusters (AuNCs) consisting of several Au atoms are chemically stable and exhibit bright fluorescence ranging from the visible to the infrared depending on the number of Au atoms in the cluster. In this study, we intend to synthesis near-infrared fluorescent gold clusters in a one-pot strategy by simply reducing a mixture of multidentate ligand and gold salt with sodium borohydride in aqueous solvent. The multidentate chelating effect affordrd by multidentate ligand suggest monodispersity and excellent colloidal stability. Then coating or encapsulating AuNCs with lipid to construct a hybrid liposome-AuNCs system to increase stability and biocompatibility. Liposomes were modified by tumor-targeted peptide for using in diagnosis of specific tumors. This study will provide a novel method for cancer diagnosis by using inorganic fluorescence nanomaterials.
