中文摘要: 钙结合蛋白介导的Ca2+信号通过调控骨组织的吸收和重塑,在肿瘤骨转移过程中发挥着至关重要的作用。本项目拟以肺癌骨转移的临床样本为研究对象,利用二维凝胶电泳和质谱技术筛选鉴定出钙结合蛋白家族的新成员钙磷蛋白CAPS1作为肺癌骨转移新的分子标志物,并进一步研究CAPS1在肺癌骨转移过程中的功能,利用体外细胞迁移实验、侵袭实验和肺癌骨转移小鼠模型分析CAPS1对肺癌细胞体外侵袭能力和体内转移能力的影响,阐明CAPS1影响肺癌细胞骨转移的分子机制。通过对CAPS1在肺癌骨转移过程中的功能及作用机制的深入研究,将增强人们对于肺癌骨转移发生机制的认识,以期确立CAPS1作为肺癌骨转移的治疗新靶点,为临床肺癌骨转移的诊断和治疗提供新的思路。

英文摘要: The calcium-blinding protein-mediated Ca2+ signal plays a crucial role in the process of bone metastases by regulating absorption and remodeling of bone tissue. This project studies the clinical samples of lung cancer with bone metastases and uses two-dimensional gel electrophoresis and mass spectrometry to screen and identify CAPS, the new member of calcium-binding proteins, as the new molecular markers in bone metastasis of lung cancer. Furthermore, it studies the function of CAPS in the process of lung cancer with bone metastases and analysis the capacity of CAPS for lung cancer cells in vitro invasion and vivo metastasis and elucidates the molecular mechanisms of the CAPS in bone metastasis of lung cancer. The intensive study for the function and mechanism of action of CAPS in bone metastasis of lung cancer will strengthen the awareness for the mechanism of lung cancer with bone metastasis, which may be established CAPS as a new treatment target for lung cancer with bone metastasis, and provides the new ideas for diagnosis and treatment in clinical lung cancer with bone metastasis.
