中文摘要: 棉铃虫性信息素的合成是由性信息素合成激活肽(Pheromone Biosynthesis Activating Neuropeptide,PBAN)调控的。目前对棉铃虫性信息素合成分子机制的研究还鲜有报道,仍需进一步的探索。本课题组前期研究发现PBAN主要通过调控脂肪酸合成途径中乙酰辅酶A羧化酶(Acetyl-coA carboxylase,ACCase)的活性来调控性信息素的合成,但是具体作用的分子机制尚需深入的研究。本研究采用RACE、Real-time PCR 、Western blot 、RNAi、GC/MS、酶抑制剂分析等分子生物学手段,以农业害虫棉铃虫为研究对象,对ACCase 的分子特性及在性信息素合成中的分子功能进行深入的研究,进而揭示PBAN对ACCase的调控机制,为解析棉铃虫性信息素合成的分子机制奠定坚实的理论基础。同时可为害虫防治寻找新的靶标,为防治奠定基础。

英文摘要: Pheromone biosynthesis activating neuropeptide (PBAN) regulates sex pheromone synthesis and release in Helicoverpa armigera. The molecular mechanism of sex pheromone synthesis regulated by PBAN still keep elusive until now and need to further study. Our previous study found that PBAN regulates the sex pheromone synthesis by mediating acetyl-coA carboxylase (ACCase) activity in the fatty acid synthesis pathway. However the molecular mechanism of PBAN- regulated ACCase activity still need to further investigate. In present study the molecular characteristics and corresponding roles of ACCase in sex pheromone synthesis are investigated by RACE, Real-time PCR, Western blot, RNAi, GC/MS, inhibitor analysis methods and so on using H.armigera as a model. These results will further reveal the molecular mechanism of PBAN-regulated ACCase, provide a foundation for elucidating sex pheromone synthesis of H.armigera and seek new target for pest control.
