中文摘要: 光动力疗法(PDT)是一种新的有效的肿瘤治疗方法,在体表肿瘤和腔道肿瘤等方面被广泛研究和应用。但由于PDT的基本元素激光穿透深度及照射范围的限制,该项技术在血液系统肿瘤领域几乎未涉足。本项目针对血液系统肿瘤病变细胞遍及循环系统的特点,提出光动力血液净化法(PDBP)这一新的治疗手段,通过类似血液透析的改良光动力疗法,以白血病为代表研究它对血液系统肿瘤的杀伤作用及免疫效应机制。项目通过细胞实验获得PDT对白血病细胞的杀伤效果和量效关系;建立大鼠急性粒白血病模型,通过血象分析、存活率等观察本项目提出PDBP对白血病病程各阶段的治疗效果,同时观察PDT后相关细胞因子的表达及细胞免疫的变化,以此分析PDT抗肿瘤免疫效应;PDT增加肿瘤免疫原性是诱导抗肿瘤免疫的基础,观察PDT后热休克蛋白和survivin的表达水平的变化,初步揭示其机制。本项目希望能为PDT在血液系统肿瘤的研究提供方法和基础。

英文摘要: Photodynamic therapy is a new effective method for tumor treatment,which has been used widly in varied tumor treatment and study such as tumor in skin and cavity.However,we almost cann't find PDT in the area of hematological malignancy as the limitation of laser penetration depth and illumination range.To aim directly at hematological malignancy,we suggest a new therapy named photodynamic blood purification method(PBPM),which simulate the idea of hemodialysis, replace dialysis semi-permeable membrane with the laser irradiation chamber to study its injurious and immune effect on leukemia,the representative of hematological malignancy. The killing effection and dose-effect relationship of photodynoc therapy on leukemia cell lines are observed by testing the survival rate of cells in vitro; The animal model of Brown Norway rat acute myelocytic leukemia(BNML) is used as experimental subjects. The therapeutic effects of PBMP on the several stages of leukemia pathogenesis curse are measured,while the relative cytokine expression and the cellular immunity changes are also detected to analysis the antitumor immunological effect.
