中文摘要: 2型糖尿病可引起高血压,但具体机制尚不完全明确,其发生可能与交感神经活性( sympathetic nerve activity,SNA)增加有关。已有研究报道瘦素可能参与糖尿病和高血压相互影响机制之中,输注瘦素可使多部位SNA增加,但瘦素对2型糖尿病压力感受性反射功能(baroreflex function)影响如何不清楚。PVN作为瘦素在中枢系统作用靶点之一,通过不同信号转导影响交感活性,本课题旨在研究2型糖尿病大鼠交感神经活性及压力感受性功能变化的基础上,对侧脑室输注瘦素,观察其对2型糖尿病大鼠交感神经活性及压力反射敏感性的影响,最后研究几种可能因子包括下丘脑黑皮质素受体3/4(MC3/4R)、神经肽Y受体1(Y1-R)、谷氨酸受体是否在室旁核(paraventricular, PVN)参与瘦素发挥影响2型糖尿病大鼠交感神经活性及压力反射敏感性的机制。

英文摘要: Type 2 diabetes can induce hypertension, but the mechanism is incompletely understood. its occurrence may be associated with increasing sympathetic nerve activity. Evidence indirectly suggests that leptin is closely related to diabetes and hypertension, infusion leptin can increase SNA in multiple organs, but the alteration of leptin to regulate the baroreflex fuction in type 2 diabetes is unknown. PVN, as one of the target sites in central nervous system, that control leptin works through different signals transduction affect the sympathetic activity. This found is based on the change of sympathetic nerve activity and baroreflex function in type 2 diabetic rats, and then intracerebroventricular leptin infusion to observe its effect on sympathetic activity and baroreflex sensitivity in type 2 diabetic rats, finally, to investigates several factors include the hypothalamus melanocortin receptor 3/4 (MC3/4R), neuropeptide Y receptor 1 (Y1-R) and glutamate receptor, which would be involved in the mechanism of leptin in PVN to change sympathetic activity and baroreflex sensitivity in type 2 diabetic rats.
