中文摘要: 哺乳动物大脑发育过程中,假复层神经上皮细胞在分裂增殖时,随着细胞周期时相的改变,细胞核呈现上下移动的现象(interkinetic nuclear migration,INM)。这种移动的意义在于能够最大可能地为分裂中的细胞提供空间,保证同一时间单位面积内的神经上皮可产生最多的子代细胞,在大脑的进化过程中具有重要意义。但是INM对于端脑发育过程中前体细胞增殖、分化的影响依然不明了,调控INM的分子机制也有待深入研究。我们前期工作中发现Forkhead转录因子家族中的成员FoxM1强烈表达于端脑的增殖区,条件性敲除FoxM1后,初步结果发现上皮细胞的细胞核不能下移至apical面,并有可能最终影响皮层神经元的分化和定位,INM受到影响。本项目将在此基础上,在端脑发育过程中,条件性敲除FoxM1,深入研究FoxM1调控INM的机制,明确FoxM1如何通过影响INM进而调控端脑细胞的增殖和分化。

英文摘要: During the development of mammalian telencephalon, a remarkable feature of neurogenesis is the apical-basal nuclear migration of the neuroepithelial cells which called interkinetic nuclear migration (INM). The movements are synchronized with the cell cycle such that nuclei move basally during G1-phase and apically during G2-phase. INM may play important roles during cortex evolution. However, it is unknown how the INM is regulated and what its effects on cell division and differentiation. Previously, FoxM1, a member of the Forkhead transcript factor family, was detected to be strongly expressed in the ventricular zone(VZ). By conditional disruption of FoxM1 in the developing telencephalon, we have found the nuclear migration to the apical lumen is slowed down. This proposal will lead to a better understanding of the function of FoxM1 in INM, cell division and differentiation by conditionally knockout FoxM1 during early development of the telencephalon.
