中文摘要: 黄酮类化合物的抑菌机制一直是研究热点。二氢杨梅素是显齿蛇葡萄(俗名藤茶)的主要活性成分,研究证明其有一定抑菌作用,有望开发成为安全、高效、低毒的天然食品防腐剂,但由于其抑菌机制一直未见相关报道,大大限制了其开发和利用。本项目在前期研究成果的基础上,以大肠杆菌和金黄色葡萄球菌为研究菌株,拟通过扫描电镜与透射电镜观察、呼吸代谢抑制实验、SDS-PAGE 蛋白谱带变化,4',6-二脒基-2-苯基吲哚(DAPI)荧光染色法、DNA超螺旋解旋、基因芯片杂交等方法,从菌体的表面形态结构、内部超微结构、细胞膜的渗透性、糖代谢途径、核酸与蛋白质的合成、DNA拓扑异构酶活性以及基因组转录水平等方面进行系统研究,从分子生物学角度和基因水平全面探索二氢杨梅素的抑菌机制,以期获得二氢杨梅素抑菌的作用靶点和关键控制点,为提高二氢杨梅素的抑菌性能及其定向改性提供理论依据,为开发高活性的二氢杨梅素防腐剂提供技术支撑。

英文摘要: The antimicrobial mechanism of flavone compounds has been a research focus.Dihydromyrecetin is the main active ingredient of Ampelopsis gro -ssedentata,many studies have investigated the effect of the antimicrobial activities of dihydromyrecetin and it was expected to exploiting the safe,efficient, low toxic natural food preservatives,but the antimicrobial mechanism of dihydromyrecetin has not seen the related reports until now,so it has greatly limited the development and utilization of dihydromyrecetin .The project based on the results of preliminary studies,in order to obtain the antimicrobial target points and critical control points of dihydromyrecetin,the Escherichia coli and Staphylococcus aureus strains were choosed to research. The antimicrobial mechanism of dihydromyrecetin was extensively investigated from the molecular biology and the gene level. It was carried out by utilizing scanning and transmission electron microscope, analyzing the respiratory metabolism,changing of the SDS-PAGE protein spectrums,4'6-diamidino-2-phenylindole (DAPI) staining assay,the inhibition of the activity of DNA topoisomerase and gene transcriptional expression.The results could provide theoretical basis for the improving of antimicrobial properties and directional modification of dihydromyrecetin,and provide technical sup
