中文摘要: 方剂配伍是中医方剂学科的核心及关键科学问题。然而,以往的中医方剂配伍研究多停留在饮片、或中药单一成分层面,定量评价中药组分配伍拮抗或协同效应较少。本项目拟开展丹参与红花活性组分配伍抗脑缺血作用规律研究,制备局灶性脑缺血再灌注损伤模型,核影像学18F-FDG PET监测活体脑组织的葡萄糖代谢,比色法测定血清中NO、iNOS及SOD,Western blot检测脑细胞内及细胞核Nrf2蛋白,RT-PCR检测脑细胞内P62蛋白及IL-1,免疫组化测定脑组织TLR4及NF-κb蛋白,ELISA法检测脑组织LXA4等,以上表征药效变化。通过数学多元逐步回归法拟合模型与计算研究,定量评价丹红组分配伍关系,阐明组分配伍规律,筛选丹红最佳配伍;探讨丹红组分配伍抗脑缺血损伤的作用机制。逐步实现中医方剂有效组分配伍研究定量化,为方剂配伍现代基础研究提供一定的思路与方法,优化临床给药方案,指导创新药物开发。

英文摘要: Prescription compatibility is the core and key scientific problems in the traditional Chinese medicine (TCM) study. However, TCM prescription study just is in pieces, or a single component level, which is short of the scientific and quantitative evaluation method of component compatibility antagonistic or synergistic effect. We aim to discover the law of function of the activity component compatibility of Salvia miltiorrhiza and Carthamus tinctorius anti ischemic cerebral diseases in this project. In the present study, we test SOD、and NO in serum of the rats with colorimetry method. We detected IL-1β and P62 mRNA levels in brain tissue with RT-PCR, and the expression of TLR4 and NF-κb immunoreactivity with immunohistochemical method, and glucose metabolism change with 18F-FDG PET, and the expression of LXA4 with ELISA method, and the expression of Nrf2 with Western blot method. We deal with the efficacy above methioned by mathematic model and a calculation method. We aim to analize comprehensivly the compatibility relation and function principle of Dan-hong component compatibility, and to summarize the regularity of compatibility, and finally to confirm the best effective component compatibility, so as to provide the quantitative study and the ideas and methods of reference for TCM prescription study. In other hand, we aim to provide optional and reasonable dosage regimen for clinical treatment so as to develop innovative drugs against the cerebral ischemia diseases.
