中文摘要: 头颈鳞状细胞癌是世界范围内第六大常见恶性肿瘤,而喉鳞状细胞癌是其最常见的类型。复方中药在抗肿瘤的同时,具有扶正祛邪、增强自身抵抗力的作用,近年来在抗肿瘤的药物研究中成为关注的热点。前期研究发现,固本抑瘤Ⅱ号对喉癌细胞的凋亡及信号通路的改变有一定的作用。为了进一步明确固本抑瘤Ⅱ号的作用及其机制,本项目拟应用喉癌细胞系和喉鳞癌原代细胞以及动物模型,验证固本抑瘤Ⅱ号抑制喉癌细胞恶性增殖、调控细胞周期的作用;应用基因表达谱芯片分析与固本抑瘤Ⅱ号疗效相关的靶向基因、信号通路及信号通路之间的相互作用,并建立分子网络;组织标本回顾性分析不同喉癌分期病人标本中靶向基因及靶向蛋白的表达水平及定位,分析靶向基因与喉癌病理参数(肿瘤大小、分期、细胞分化程度、淋巴结转移、远处转移)及预后的相关性,阐明固本抑瘤Ⅱ号对喉癌的作用机制,探讨中医药作为中晚期喉癌综合治疗手段之一的新策略。

英文摘要: Head and neck squamous cell carcinoma (HNSCC) is the sixth most frequent cancer, and laryngeal squamous cell carcinoma (LSCC) is the most common type.Compound traditional Chinese medicine has become the focus of attention in the research of anti-tumor drugs as it can be antineoplastic as well as strengthening the body resistance to eliminate pathogenic factors.Previous study has found that Guben Yiliu II (GY II) could promote the apoptosis of laryngeal carcinoma cells and change the signal pathways.In order to clarify the effect and mechanism of GY II,the following work should be compeleted.Primary laryngeal carcinoma cells and Hep-2 cell line and animal models will be used to verify the effect of GY II on inhibiting malignant proliferation and adjusting the cell cycle; Microarray gene expression analysis will be used to find the targeted genes, the signal pathways and the interaction between the pathways related to GY II,and a network of the moleculars will be tried to created;Different clinical stage patient specimens will be analyzed retrspectively to find the location and the expression levels of targeted genes and proteins;The correlation between target genes and laryngeal pathological parameters (tumor size, stage, cell differentiation, lymph node metastasis, distant metastasis ) and prognostic relevance will be also analyzed.The objective is to verify the mechanism of GY II ,and investigate the laryngeal squamous cell carcinoma therapeutic strategies concerning traditional Chinese medicine.
