中文摘要: 天然产物历来是药物发现的重要源泉之一。来源于旋花科植物的树脂糖苷类化合物,因结构独特新颖、生物活性多样,部分相关植物药食同源,近年来受到了极大的关注。鉴于该类化合物存在的分离困难、构效关系及作用机制研究不明确的问题,本课题组建立了利用LC-TOF-MS的化学结构导向发现技术,并结合成分和结构-活性和机制-中药和功效三者相关的CAT数据库信息进行新树脂糖苷类化合物快速发现的方法,并在前期研究中发现了包含多个新颖骨架的一系列树脂糖苷类成分,且活性研究结果显示该类化合物具有较强的逆转肿瘤MDR和抗α-溶血毒素活性。本课题拟在前期研究基础上,从药用植物丁香茄、菟丝子等五种药用旋花科植物中寻找和发现结构丰富多样的树脂糖苷类成分,并系统研究该类化合物逆转肿瘤MDR和抗α-溶血毒素作用,阐明其构效关系及作用机制。

英文摘要: Natual Products played an important role in the history of drug discovery. Resin glycosides, mainly found in Convolvulaceae, gained more and more attention because of their intriguing structure, diverse activity and edible plant resource. Considering the uncleared QSAR and pharmacological mechanism of the resin glycosides, together with their difficulty in separation, we established an quick and guided method to discover novel resin glycosides from Convolvulaceae based on LC-TOF-MS analysis and CAT data, which led to the isolation of a series of resin glycosides with scarce skeleton. These resin glycosides showed significant potency of multi-Drug resistance reversal activity and α-hemolysin inhibition in our previous research. Based on these studies, we expect to get various structural resign glycosides from Convolvulaceae, and futher investigate their multidrug resistance reversal activity and anti-α-hemolysin activity, as well as their QSAR and pharamacological mechanism.
