中文摘要: 疾病诊断关键是对疾病标志物进行检测,发展疾病标志物及相关信息的高灵敏检测方法具有重要的科学意义和应用前景。本项目以分析化学与分子生物学、纳米技术和材料化学交叉研究为主旨,以几种肿瘤标志物(如P53蛋白、癌胚抗原、Bcl-2抗原、人半乳糖凝集素-7、急性白血抗原等)的传感检测为切入点,重点开展几种标志物检测新方法的研究。利用上转换纳米粒子在红外光照射下发可见光,消除生物检测背景,提高灵敏度,开发高灵敏的肿瘤标志物检测技术;结合上转换纳米材料发射波长可调的优势,通过调变原料组成、合成条件等获得同一激发波长下发射不同波长荧光(红、绿和蓝光)的上转换纳米材料,用其对肿瘤标志物抗体编码,建立多种肿瘤标志物的同时检测方法;利用编码探针对肿瘤细胞进行识别,实现对肿瘤细胞的可视化检测。本课题的研究可望发展肿瘤标志物以及肿瘤细胞生物分析的新原理、新方法和新技术,对重大疾病的早期诊断与预警具有重要意义。

英文摘要: The detection of disease markers is the key of clinical diagnosis. The development of highly sensitive detection methods for diseases markers are of critical importance and offer great application prospects.This program is forcused on the development of new biosensors and detection strategies for tumor markers(e.g. tumor protein p53, carcinoembryonic antigen, Bcl-2 antigen, human galectin-7, and acute leukemia antigens) analysis based on the interdisciplinary research of analytical chemistry, molecular biology, nanotechnology and material synthesis. Upconversion nanoparticles with visible radiation under near-IR irradiation will be systhesized and used to eliminate the background signals and to enhance the sensitivity of the target biological assay systems in this program.By verying the components or preparation conditions, the synthesized upconversion nanoparticles altered within emitting red, green, and blue light at the same exitation wavelength.With the application of the novel upconversion nanoparticles, it is realizable to encode the antibodies of the tumor markers and obtain encoded specific nanoparticle detecting probe for tumor markers.By using the encoded detecting probs, a novel tumor marker array will be established.
