中文摘要: 银屑病关节炎(psoriatic arthritis, PsA)致残率高、危害大,早期预测和诊断是阻止其进程、改善预后最有效的方法。但是,目前临床上没有成熟的生物标志物可用于PsA的早期预测和诊断,已成为有效控制其发生发展的瓶颈。MicroRNA调控基因的表达,是用于疾病进程预测的理想生物标志物。本项目将在前期研究的基础上进一步开展以下研究:1)对中国人群PsA患者的血清microRNA特异性生物标志物进行研究,筛选出早期诊断的生物标志物;2)通过对银屑病进展到PsA前后的血清样本的动态采集并进行microRNA表达谱研究,筛选早期预测的microRNA生物标志物;3)依据中医对PsA寒热证型的分类,探讨PsA寒热辨证与血清microRNA生物标志物的关系。本项目将对PsA的早期诊断、预测和早期干预提供生物学依据,并将为中医辨证分型的规范化奠定基础,具有重要的科学意义和临床价值。

英文摘要: Psoriatic arthritis (PsA) can lead to joints damage, with a high disability rate. The early diagnosis and treatment are the best methods for preventing PsA disease process and improveing PsA prognosis. However, there is no specific biomarkers for prediction and early diagnosis of PsA, which is the bottleneck for effective control of this disease. MicroRNA is able to regulate the expression of genes, and thus can be the perfect biomarkers for prediction of disease progression. This project intends to perform the following studies: 1) Screening out the early diagnosis biomarkers by the study on serum specific microRNA in Chinese population; 2) Exploring the early predictive microRNA biomarkers through detecting the serum samples of PsA patients, premorbid samples as well as the dynamic acquisition samples from psoriasis vulgaris to PsA;3)dertermination of the discriminating cold syndromes from hot syndrome using the serum microRNA biomarkers tested. This study will provide the biomarkders for the early prediction, early diagnosis and early intervention of PsA, and it is also helpful for the standardization of syndrome differentiation for Chinese medicine.
