中文摘要: 核技术在小麦遗传改良和品种选育中取得了显著成效,开展诱发突变机理研究有助于指导和促进核技术在农业生产中的应用。核辐射所致DNA损伤是诱发突变的根源,其中DNA双链断裂(DSBs)是辐射最重要的原初损伤。由Ku70和Ku80组成的异源二聚体Ku蛋白,在DNA损伤修复途径中发挥识别DSBs和募集下游修复因子的核心功能。动物细胞和模式植物中Ku70和Ku80基因的表达都与辐射敏感性相关。本项目在六倍体小麦中成功克隆了TaKu70和TaKu80基因,初步分析表明TaKu70和TaKu80具有Ku蛋白的保守特征。本项目拟通过蛋白结构域和功能分析、亚细胞定位,以及拟南芥转基因研究TaKu70和TaKu80的功能;通过不同小麦品种中TaKu70和TaKu80对伽马射线的应答模式分析,研究其对辐照损伤的应答机制和与辐射敏感性间的相关性。

英文摘要: Error-prone nature of DNA damage-repair can induce mutation that can potentially be transmitted to the progeny.Ku protein, which is a heterodimer consisting of the Ku70 and Ku80 subunit, is of central importance to DNA damage-repair and shows high correlation with radiosensitivity. So it can be the optimal starting point to study the mechanism of radiation induced mutagenesis in wheat and other main crops.Our group has cloned the cDNA sequenses of TaKu70 and TaKu80 genes in wheat. Sequense analysis shows deduced amino asids sequenses of these genes contain highly conserved domains of Ku protein in higher eukaryotes, which imply the function in DNA damage-repair. In this proposal, we plan to study the main characteristics of TaKu70 and TaKu80 genes, including gene structure, chromosomal localization, phylogenesis, spatial and temporal expression profile, subcellular localization and the biochemical functions of deduced polypeptids by using molecular approachs. We propose to clarify the in planta function of TaKu70 and TaKu80 genes in DNA damage-repair and growth regulatin by using over expression and RNAi transgenic approachs. Then we will study the radiation response models of TaKu70 and TaKu80 genes in different wheat cultivars, in order to analysis their correlation with wheat radiosensitivity.
