中文摘要: 流感病毒感染致病涉及病毒与机体两个复杂生物系统及其二者间的相互作用。一方面,宿主免疫细胞识别病毒感染并诱导产生I型干扰素以清除病毒;另一方面,病毒亦形成多种机制逃避机体免疫系统识别和攻击,逃逸天然免疫的监控清除,从而感染机体。流感病毒激活的宿主RIG-I信号途径在机体免疫和病毒逃逸方面发挥着重要作用。本课题采用Real Time RT-PCR技术、免疫共沉淀法、ELISA技术,观察甲型流感病毒感染基因敲除小鼠腹腔巨噬细胞和基因敲除小鼠肺脏中病毒载量、RIG-I、NF-κB、MAVS、IRF3、Siglec-G、SHP2、c-Cb1蛋白活性,以及细胞因子表达,从细胞、分子和基因水平阐明流感病毒感染的生物学机制,并在此基础上探讨金柴抗病毒胶囊抗病毒以及干预病毒免疫逃逸作用靶点,为揭示中药整体调节的抗病毒机理提供实验依据。

英文摘要: Pathological reaction of virus infection disease involve in virus and host immunity.The immune cells induced type I interferon to clear virus; but the virus has also formed a variety of mechanisms to against the body's immune system,to infect the body and cause chronic infection ,The host signal pathway of RIG-I activated by the influenza virus plays an important role in immune and antiviral escape aspects. we will use Real Time RT-PCR to detect viral load expression in lung from gene knockout mouse and peritoneal macrophage,use coimmunoprecipitation to detect RIG-I, NF- κB, MAVS, IRF3, Siglec-G, SHP2, c-Cb1 activity, and use Enzyme-Linked Immunosorbnent Assay to detect expression cell factor. The aim of this study is to explore the antiviral targets of jin Chai antiviral capsule , and to provide experimental basis for revealing the antiviral mechanism of Chinese herbal medicine regulating.
