中文摘要: 目前,研究表明鸡传染性法氏囊病毒(IBDV)感染机体后能够引起机体I 型干扰素(IFN)下调从而抑制机体的先天性免疫应答,但是究竟是IBDV的哪个蛋白参与引起机体I型IFN的下调,并未做深入研究。申请人首次用Luciferase 试验等证实IBDV VP4和VP5蛋白能够下调机体I 型IFN产生的信号通路和发挥效应的信号通路。在此基础上,本项目将进一步从RIG-I 或IPS-1介导的IFN 信号通路、IRF3 信号通路、NF-κB信号通路等IFN产生的信号通路上阐明VP4和VP5蛋白引起机体I 型IFN 下调的分子机制;从IFN 发挥效应的信号通路上,尤其是JAK-STAT 信号通路来阐述IBDV VP4和VP5影响I 型IFN 发挥效应的机制。由此,本项目将系统地阐明IBDV的VP4和VP5蛋白影响机体I 型IFN 信号通路的分子机理,为预防和控制IBDV感染提供新的靶向分子。

英文摘要: Infections bursal disease virus (IBDV) infection could down-regulate type I interferon (IFN) to disarm host innate immune response, but the proteins of IBDV which had this function had never been studied. In this study, it was confirmed that VP4 and VP5 of IBDV could inhibit type I IFN production and signaling pathway using luciferase assay. The mechanisms of VP4 and VP5 of IBDV down-regulating type I interferon would be further studied from the RIG-I or IPS-1-mediated IFN signaling pathway, IRF3 signaling pathway, NF-κB signaling pathway. From IFN-mdediated JAK-STAT signaling pathway, the mechanism of VP4 and VP5 of IBDV affecting the effect of IFN was further explored. Therefore, the project will systematically study and elucidate the mechanism of VP4 and VP5 of IBDV inhibiting the type I IFN signaling pathway, thus it will provide new target molecules for the prevention and control of IBDV infection.
