中文摘要: 日本血吸虫病仍然严重危害人类健康。对其感染免疫应答及其调节机制缺乏认识是目前血吸虫病疫苗研究的重要瓶颈。本课题组近期研究发现日本血吸虫感染引起宿主脾脏淋巴细胞凋亡是其导致宿主免疫抑制的重要原因之一,进一步预实验表明虫卵抗原致脾脏T淋巴细胞凋亡作用显著。本研究拟首先筛选鉴定致小鼠脾脏T淋巴细胞凋亡的虫卵分子(群),通过对其致凋亡通路及原因分析探讨日本血吸虫感染致小鼠脾脏T淋巴细胞凋亡的机制,并观察目的分子(群)作为疫苗的抗感染及抗病作用。本研究的开展一方面将为深入研究日本血吸虫感染致宿主免疫抑制或下调机制奠定基础,另一方面为解释日本血吸虫致病机制以及复杂的宿主-寄生虫互作关系提供新思路。

英文摘要: Schistosomasis japonicum remain a serious threaten to public health. Insufficient understanding of the mechanisms of immune response and its immunoregulation restricted vaccine development. Our previous study showed that apoptosis of splenic lymphocytes induced by Schistosoma japonicum contributed to down-regulate host immune responses during infection, and we resently observed that antigen from eggs has the most significant pro-apoptotic acticity. In this study, we will firstly sreen and identify molecule(s) from Schistosoma japonicum eggs with pro-apoptotic effect to splenic T lymphocytes, and then explore the underlying mechanisms by clarification of the apoptotic signal pathway. This study will make a foundation for the future research of host immuno-dowm regulation in schistosomasis japonica and provide novel candidate molecules for study on anti-infection vaccine. It will benefit for explanation of the pathology of schistosomasis japonica from the molecular aspect and illuminating of the complicated host - parasite interaction.
