中文摘要: 系统性红斑狼疮(SLE)是一种累及全身多系统、多脏器的自身免疫性疾病,其发病机制目前还不明确。中医学临床辨证将SLE主要分为3个证型,其中热毒炽盛证是SLE临床常见证型之一。最新的研究表明,微小RNA(microRNA,miRNA)在多种疾病,包括自身免疫病的发生、发展中起着非常重要的调控作用。本课题以SLE中医临床辨证常见证型热毒炽盛证患者为研究对象,以SLE脾肾阳虚证患者和阴虚火旺证患者为阳性对照,正常人为阴性对照,利用生物信息学miRNA芯片技术和分子生物学技术,研究SLE热毒炽盛证患者miRNA的差异表达及其功能,找出SLE热毒炽盛证具有特异性的差异表达miRNA,并研究其发挥作用可能的机制。本课题将现代生物信息学技术、分子生物学技术和传统中医辨证的证型结合起来探讨SLE的发病机制,从而可能为SLE热毒炽盛证提供新的现代生物医学方面的理论支持。

英文摘要: Systemic lupus erythematosus (SLE) is an autoimmune disease that involved in multiple organs and systems. Up to now,its pathogenesis has not yet been clear.In Clinical,TCM classifies SLE into three syndrome types, in which, toxic heat flaming syndrome is a common one. The latest research showed that microRNA has very important role in regulating a variety of diseases and their development, including the autoimmune diseases. In this study, we select the common SLE TCM syndromes, the toxic heat flaming type as the research object.Patients with syndrome of yang deficiency of spleen and syndrome of hyperactivity of fire due to yin deficiency serve as positive controls.The normals serve as negative controls. We use bioinformatics and molecular biology techniques to study the toxic heat flaming syndrome in SLE patients. MicroRNA differencal expressions and function as well as microRNA target genes will be investigated. The possible role of microRNA and the mechanisms will be explained. Our study combines modern bioinformatics and molecular biology techniques with TCM syndromes, SLE toxic heat flaming syndrome, which is to explore the pathogenesis of SLE and may provide a new aspect of modern biomedical and theoretical support.
