中文摘要: 前体mRNA(pre-mRNA)的可变剪切是真核基因表达调控的重要方式。大部分人类基因都有前体mRNA的可变剪切。很多研究都表明可变剪切在癌症的发生发展中起了重要作用。前体mRNA剪切方式的异常改变可能产生或上调一些致癌性蛋白,或者失活抑癌基因,而有利于癌症的发生发展。核内不均一核糖核蛋白C(hnRNP C)是重要的可变剪切调控蛋白。研究表明hnRNP C与肿瘤发生和转移有关。本项目在前期研究的基础上提出hnRNP C可能在口腔黏膜异常增生演变中起了重要作用。我们将采用免疫组织化学、荧光定量PCR、RNAi、裸鼠和转基因鼠等实验方法,研究hnRNP C在口腔黏膜异常增生和肿瘤组织中的表达模式和作用,揭示hnRNP C参与口腔黏膜异常增生和口腔癌的发生发展的机制,为口腔黏膜异常增生、口腔癌及其它癌症的治疗提供新的标靶和思路。

英文摘要: Alternative splicing of pre-mRNA is an key regulation way of the expression of eukaryotic genes. Most of human genes undergo alternative splicing of pre-mRNA. More and more evidences indicate that alternative splicing plays key roles in the initiation and developement of cancer. Alternative splicing may participate in cancer by increasing the expression of oncogenes, producing oncogenic unusual proteins, or deactivating tumor suppressor genes.hnRNP C is an important splicing factor.It has been shown that hnRNP C is involved in the development and metastasis of cancer. According to our pilot studies, we proposed that hnRNP C may play important role in the progress of the dysplsia of oral mucosa. The goal of this proposal is to undercover the functions and mechanisms of controling the initiation and progress of oral mucosa dysplasia by hnRNP C. This study may find new targets or ways for the treatment of oral cancer and oral mucosa dysplasia.
