中文摘要: 海洋有害微生物是影响国民经济建设的重要因素之一,开展微生物检测和控制研究对国民经济建设有重大意义。本项目拟以寡糖活性小分子作为功能单元,在有机合成化学基础之上,结合特定的纳米材料,在单细胞水平上检测和分析海洋微生物。项目主要内容为:重点设计功能化的糖类分子(N-乙酰胞壁酸、N-乙酰葡糖胺、L-甘油-D-甘露-庚糖和棉籽糖)分别与量子点、磁性纳米材料和荧光素交联形成活性小分子功能纳米探针,考察这些小分子功能纳米探针在微生物快速检测和微生物膜原位表达分析中作用规律。本项目创新性体现在:分子水平上揭示活性小分子探针功能化的纳米材料与微生物作用关系,微观尺度水平上总结功能活性小分子与微生物膜相互作用的规律,特别为解决涉及微生物膜内"时间"、"空间"和"动态"变化的复杂生物学问题提供参考和借鉴。

英文摘要: The detection of pathogenic bacteria is key to the prevention and identification of problems related to health and safety. Legislation is particularly tough in areas such as the marine engineering, where failure to detect an infection may have terrible consequences. This project aims to research the nanoprobe combined with the saccharide molecules as functional elements and nanomaterials as response lables in marine microbial detection and analysis in the single cell level. The oligosaccharide-based nanoprobes including N-Acetylmuramic acid,N-Acetylglucosamine,L-glyero-D-manno-heptose,and raffinose,conjugated with quantum dots, magnetic nanomaterials and fluorescent,respectively, will be devised. The performance of nanoprobe in bacteria detection and the functional express of biofilm will be studied. Despite these remaining challenges, a very bright future of small molecule nanoprobes can still be expected as a result of the presence of huge development space for researchers to better settle the issues mentioned above and explore more novel applications by designing and synthesizing novel nanoprobe. Looking ahead, we speculate that small molecules nanoprobes will display functions in more sophisticated and more in-depth biological systems, specially, the "spatiotemporal" and "dynamic" changes in complex biofilm.
