中文摘要: 由于抗寒高产的优良苜蓿品种短缺,在很大程度上限制了我国北方寒冷地区苜蓿的种植及其草产业的发展,发掘和利用抗寒植物基因资源,加快抗寒苜蓿新品种培育是解决该问题的重要途径。扁蓿豆是苜蓿属多年生二倍体植物,具有抗寒、抗旱、抗蓟马等优良特性,是遗传改良提高苜蓿抗逆性的理想材料。 本项目以直立型扁蓿豆为主要研究对象,在大田自然环境和人工控制的低温胁迫条件下,比较分析扁蓿豆不同生长状态下各器官形态结构、养分累积规律及与逆境胁迫相关的生理生化指标的变化。利用已知蒺藜苜蓿的遗传背景,研究已克隆的3个扁蓿豆抗寒相关基因的时空表达特性及在不同苜蓿材料上的表达差异,并分析其与各材料形态结构、生理生化变化的相关性。进一步通过遗传转化验证其抗寒功能,探讨扁蓿豆抗寒功能基因在低温胁迫下的响应机理,明确其对紫花苜蓿遗传转化后的抗寒效果,为苜蓿的抗寒高产分子标记辅助育种及转基因育种奠定基础。

英文摘要: The development of alfalfa planting and grass industry in the northern cold regions were largely limited, due to the lack of excellent alfalfa varieties which have strong cold resistance and high yield. It is an important way to solve the problem that explore and use of plant genetic resources have cold resistance and accelerate the cultivation of new varieties of alfalfa. Medicago ruthenica is a perennial legume diploid plant in Medicago. It has excellent characteristics such as strong thrips resistance, strong cold and drought resistance. It is ideal material to improve the resistance of alfalfa for genetic improvement. In this study, Medicago ruthenica(L.)cv.Zhilixing will be taken as material. Morphology, nutrient accumulation and physiological and biochemical changes related stress will be comparative analysised in the field and cold stress. Temporal and spatial expression characteristics of the cloned three cold-related genes of Medicago ruthenica and differentially expressed in different alfalfa materials will be studied using known genetic background of Medicago truncatula ,at the same time ,the correlation with morphology, physiological and biochemical changes will be revealed . They will lay the foundation for molecular marker and transgenic alfalfa breeding on cold resistance and yield of alfalfa.
