中文摘要: 角膜的无血管状态是保持角膜透明度以及角膜免疫赦免状态的重要解剖基础,多种角膜疾病可以导致角膜新生血管的产生,严重影响患者视力。既往的研究认为,角膜内促新生血管因子与抗新生血管因子的平衡是维持角膜无血管状态的关键,在角膜损伤或炎症时,这一平衡状态被浸润的炎症细胞打破,从而产生新生血管。最近,我们以及其他团队的研究发现,炎症细胞在角膜新生血管发生中可能不发挥关键作用。在本研究中,我们将深入观察角膜基质细胞、角膜基质微环境以及基质细胞与上皮细胞的相互作用在不同原因诱导的新生血管形成过程中的动态变化,探讨基质细胞在角膜新生血管发生发展过程中的核心作用。本项目的实施将有助于我们从角膜基质细胞角度认识角膜新生血管形成的分子机制,为我们寻找角膜新生血管治疗新靶点奠定基础。

英文摘要: Corneal avascularity is an important anatomic basis for corneal transparency and immune privilege. Many corneal diseases can cause corneal neovascularization and result in severe vision loss. Previous studies have demonstrated that the balance between angiogenic factors and antiangiogenic factors in the cornea plays the pivotal role in the maintenance of corneal avascularity. During corneal wounding or inflammation, the balance will be compromised by infiltrated inflammatory cells and then cause corneal neovascularization. However, recent works from others and ours indicated that inflammatory cells may not play the key role in the process of corneal angiogenesis. In this project, we will focus on the dynamic change of corneal stromal cells, i.e, keratocyte, corneal stromal microenvironment and stromal-epithelial interaction on the corneal angiogenesis process, so as to investigate the function of keratocyte on corneal angiogenesis. Fulfillment of this project will help us to understand the molecular mechanism of corneal angiogenesis from the point of keratocyte, and set the basis for searching new target for the treatment of corneal neovascularization.
