中文摘要: 纳米光热材料研究的高速发展为精确治愈肿瘤提供了新的思路,但由于生物体内的微环境较为复杂,肿瘤细胞能够不断适应并躲避纳米材料的识别,加之肿瘤组织结构致密、缺乏淋巴回流的特点,使得纳米材料很难有效地靶向并渗透肿瘤组织发挥抗癌功效。本项目拟采用干扰素-γ (IFN-γ) 活化巨噬细胞,提升巨噬细胞免疫靶向渗透肿瘤组织以及细胞吞噬装载金纳米棒的能力,并对巨噬细胞运载金纳米棒靶向渗透肿瘤组织的输送机制进行研究。当巨噬细胞将金纳米棒运载到乳腺肿瘤部位后,金纳米棒在近红外激光的照射下将光能转换为热能,进而将乳腺肿瘤细胞杀死。本项目将巨噬细胞高靶向识别肿瘤、抑制肿瘤生长的免疫能力与金纳米棒光热治疗肿瘤相结合,克服纳米材料在体内靶向、渗透肿瘤能力的不足,降低纳米材料在非治疗区域的残留。从而构建以巨噬细胞为载体的高靶向输送金纳米棒光热治疗乳腺肿瘤的治疗策略。

英文摘要: The rapid development of nano thermal materials provide a new strategie for precisely treatment of tumor. But due to the complicated micro-environment in vivo, tumor cells adapting to avoid the identification of nano-materials, compact tissue structureof tumor and the lack of lymphatic circumfluence, it is difficult for nano-materials to effectively target and penetrate the tumor, exerting anti-cancer effects. This project aims to use interferon gamma (IFN-γ) to activate macrophages, enhancing the ability that macrophage immune targeted penetrates tumor tissue and phagocytic delivery gold nanorod, and then to study the mechanism of macrophages delivering gold nanorods. When macrophages deliver gold nanorods to the breast tumor, gold nanorods under the near-infrared laser converted light energy into heat energy, killing tumor cells. In this project, macrophages highly immunity targeting tumor synergy with gold nanorods photothermal therapy for cancer to overcome nano-materials low targeting, defects of penetrating tumor and reduce nanomaterials remain in the non-treatment area. Thus, we are to establish a breast cancer photothermal treatment of macrophage highly targeting delivery gold nanorods to tumor.
