中文摘要: 多糖是一类重要的免疫调节剂,其作用位点研究是生命科学领域的热点和难点。阿里红是新疆具有特色的维药资源,前期研究发现阿里红粗多糖(FOP)具有较好的免疫调节作用。本项目拟在前期工作基础上,以阿里红中分离的FOP为研究对象,运用化学分析和仪器分析 (GC和GC-MS,MS,NMR,HPLC-ESLD) 等现代多糖分析方法, 阐明阿里红多糖的化学结构。同时,观察其对T、B淋巴细胞和巨噬细胞活化和增殖功能的影响,确认该活性多糖免疫调节作用的靶细胞;并拟从CD19、TLR4、TLR2和 CR3等已知多糖受体为切入点,应用模式识别受体的单克隆抗体与多糖免疫细胞膜表面已知受体进行竞争性抑制,采用抗体封闭法筛选和鉴别FOP在靶细胞上可能的作用位点,在此基础上,初步探讨FOP对 TLR4/ NF-κB信号通路的调节作用,旨在部分阐明阿里红多糖免疫调节作用的分子机制,为维药多糖的免疫活性研究提供?????。

英文摘要: Polysaccharides are a kind of important immunomodulators, its receptor or binding site research are the hot and difficulty in life science. Fomes officinalis has the characteristics of the Ughur herbal resource in Xinjiang, previous study found that Fomes officinalis crude polysaccharide (FOP) has a good immunoregulation effect. Based on our previous study, in order to Fomes officinalis separation of FOP as the research object, using a chemical analysis (GC and GC-MS, MS, NMR, HPLC-ESLD) and other modern polysaccharide analysis methods, to elucidate the chemical structure of polysaccharide of Fomes officinalis. Meanwhile, Firstly, we will study its effects on the proliferation and activation of T、B lymphocytes and macrophages and identify the immune target cells. Secondly, we will screen TLR4, TLR2, CD19 and CR3 as the potential polysaccharide immune receptors on B cells and macrophages membrane with the antibody block method, and then take the gene mutant mice as the research target to verify the binding site by 3H-TdR incorporation test, ELISA and biolayer interferometry. Furthermore, we will study the TLR4/ NF-kB signal pathway of its immunoactivation.
