中文摘要: NLRP3炎性小体活化在2型糖尿病等多种炎症性疾病中发挥重要作用。NLRP3与UUO小鼠肾脏炎症、肾损伤和纤维化密切相关。目前,NLRP3炎性小体在糖尿病肾病发病中的作用还不清楚。本研究将利用基因敲除鼠(NLRP3-/-和TXNIP-/-)观察NLRP3和TXNIP在糖尿病肾损伤中的作用和分子机制,以及二者的相互作用关系。体外原代培养NLRP3-/-、TXNIP-/-和野生型小鼠肾小管上皮细胞,观察NLRP3对糖尿病状态下细胞凋亡和转分化的作用及其分子机制,以及TXNIP与NLRP3之间的相互作用。同时,利用基因转染、siRNA干扰等技术观察NLRP3与高糖诱导的人肾小管细胞凋亡和表型转化的关系以及相关分子机制。本研究将有助于揭示NLRP3在糖尿病肾病发病中的作用和分子机制,为糖尿病肾病防治提供新的作用靶点和思路,具有重要的理论意义和实用价值。

英文摘要: NLRP3 inflammsome activation contributes to the inflammatory diseases including type 2 diabetes. NLRP3 contributes to renal inflammation, injury, and fibrosis following unilateral ureteric obstruction in mice. At present, the role of NLRP3 inflammasome in diabetic nephropathy is unknown. In this project, we will investigate the effect and molecular mechanism of NLRP3 and TXNIP on diabetic nephropathy, and the interaction of both using NLRP3-/- and TXNIP-/- mice. We will observe the effect of NLRP3 on apoptosis and epithelial-mesenchymal transition in renal tubular cells under diabetic condition using primary cultured renal tubular cells from NLRP3-/-, TXNIP-/- and wild type mice. Meanwhile we will also investigate the role and moleculor mechanism of NLRP3 on HG-induced apoptosis and epithelial-mesenchymal transition in huamn renal tubular cells using gene transfection, siRNA interference and other molecular biology techniques. This project will be helpful to reveal the role and molecular mechanisms of NLRP3 in diabetic nephropathy. And this project has important theory significance and practical value for offering a new therapeutic strategies for the prevention and treatment of diabetic nephropathy.
