中文摘要: CB2(大麻素2型)受体与G蛋白体系在生物体内与镇痛机制密切相关。实验表明CB2受体激活后,与G蛋白的两类亚型Gi和Gq存在相互作用,但与Gs蛋白是否存在相互作用仍未知。本课题将发展联合残基模型UNRES膜蛋白模拟和双蛋白质链折叠动力学方法,对膜蛋白CB2受体进行分子动力学模拟,研究CB2受体和G蛋白双链折叠作用过程和机制,预测它与Gs蛋白是否偶联。这项研究不仅将为探寻外周镇痛机制研究提供新的理论依据,也将为其他蛋白质体系的研究提供新的方法和思路。

英文摘要: CB2 receptor (cannabinoid receptor 2) coupling G proteins had a strong inhibitory effect on pain in biological systems. Previous studies indicated that the active CB2 receptor coupled with the downstream of Gi and Gs proteins mediated analgesic signal pathways. However, the interaction between CB2 and Gs was not clear until now. The purpose of this proposal is to construct and test eligible force field to simulate the transmembrane protein, CB2 receptor by UNRES model, and develop dual-chain protein folding program to simulate interactions between CB2 receptor and G-protein and study the interaction mechanisms. The results of simulations will be meaningful for the further prediction of interaction between CB2 receptor and Gs protein. As a whole, this proposal will not only focus on peripheral analgesic mechanisms, but also provide new methods to study protein-protein interaction problems.
