中文摘要: 酮与氨的不对称还原胺化被认为是合成手性胺的最理想反应。而胺脱氢酶催化的该反应因更加绿色,将是未来手性胺合成研究的热点。但目前所报道的胺脱氢酶存在活性低、底物谱极窄的问题,分子改造也因对其结构功能关系不明而无法有效地进行功能强化,严重影响了其在手性胺合成中的应用。针对这一问题,本项目以筛选获得的氨基酸脱氢酶为起始对象,通过定向进化将其进化成对模式酮底物有胺化还原活性的胺脱氢酶,解析该酶及其与底物复合物的三维结构,探索酶与底物结合和催化的关键氨基酸残基,揭示酶结构变化对功能转变的影响规律,阐明其催化的分子机制;其次,利用获得的结构与功能关系,针对不同类型的脂肪族和芳香族酮底物,通过分子对接和分子动力学模拟,探索关键突变位点,对酶进行针对性的理性设计与分子改造,拓展其底物谱,针对不同底物获得相应的高效胺脱氢酶;最后,对其催化反应体系进行系统优化,为手性胺规模化生产构建一个高效绿色的生物催化过程。

英文摘要: The asymmetric synthesis of chiral amines from prochiral ketones and free ammonia is considered to be one of the top aspirational reactions.The reductive amination of ketones catalyzed by amine dehydrogenase will play a particularly important role in chiral pharmaceutical industry.Although some amine dehydrogenases have been developed,low activity and narrow substrate specificity restrict their practical application in synthesis of chiral amines.The proposed project plans firstly to investigate the relationship between structre and function of an amine dehydrogenase.Subsequently,using the protein rational design strategy to improve activity and broaden the substrate specificity of the amine dehydrogenase based on the structural information.Finally, the green and efficient biocatalytic processes catalyzed by amine dehydrogenases for the production of various chiral amines are successfully developed.
