中文摘要: 本项目旨在构建一种新型体内长循环、受体靶向、还原敏感释药及逆转肿瘤细胞多药耐药性的多功能胶束载药系统,以期提高抗肿瘤药物的治疗指数。课题设计思路是将具有长循环功能的Heparosan多糖,通过二硫键,与维生素E琥珀酸酯(VES)链接,合成两亲性共聚物载体材料Heparosan-S-S-VES,然后,创新性地选择Her-2多肽修饰,制备多功能靶向聚合物胶束给药系统,用于细胞内递送阿霉素。该研究对于拓展新型Heparosan多糖在药剂学领域的应用以及完善纳米靶向给药系统的设计具有很好的科学意义。此外,深入考察聚合物胶束的摄取机制、胞内转运、载体和/或药物在亚细胞器的分布等,可望从细胞水平上为今后抗肿瘤药物聚合物胶束系统的研究提供科学参考。

英文摘要: The main objective of the proposal is to construct a novel multifunctional polymeric micelle drug delivery system with the effects of long circulation, acceptor targeting, redox-sensitive release and reversal of multidrug resistance. Amphipathic copolymer of Heparosan-S-S-VES is synthesized and decorated with her-2 peptide to prepare polymeric micelles, which are used for intracellular targeting delivery of doxorubicin. Furthermore, the endocytosis mechanism, intracellular transport and the distribution in organelles of the micelles are researched deeply in tumor cells. The study of the project will extend the application of heparosan in pharmaceutics, and will provide scientific references for design of polymeric micelle delivery systems in the future.
