中文摘要: 缺血性心脏疾病严重威胁人类的生命安全与健康,常伴随心肌细胞的大量死亡,目前的治疗手段虽可缓解症状但却无法根治,因此迫切需要新的方法来修复受损心肌组织以重建心脏的功能。心肌细胞移植为缺血性心脏疾病的治疗带来了新的希望,但长久以来存在着细胞成活率低的问题。本申请拟针对这一问题进行应用基础研究,设计系列壳聚糖-柠檬酸-抗坏血酸缩合物,用以制备抗氧化可注射可降解水凝胶心肌细胞支架,期望获得理想的心肌细胞移植成活率。壳聚糖、柠檬酸和抗坏血酸可协同抗氧化,应对组织损伤处原有和外物引起的氧化应激;壳聚糖生物相容性极好,柠檬酸为机体固有物质,抗坏血酸为机体必须的营养素,三者结合确保了生物相容性;以注射方式传递所移植细胞入组织损伤部位,减少手术的危害与疼痛;注射后水凝胶在体温作用下呈不可流动的半固体状态,支撑所传递的细胞;随着所移植心肌细胞的良好生长,支架材料逐渐降解排出体外,最终重建心脏功能。

英文摘要: Ischemic heart disease is a serious threat to human safety and health, often accompanied by massive death of cardiomyocytes. Although current treatments can relieve symptoms, it still can not completely cured, there is an urgent need for new ways to repair damaged heart tissue to rebuild the heart function. Myocardial tissue transplantation for the treatment of ischemic heart disease has brought new hope, but there is a long-standing problem of low survival rate of the cells. This application is intended to carry out basic research to solve this problem, design series of chitosan - citric acid - ascorbic acid condensates, for the preparation of antioxidative injectable biodegradable hydrogel scaffold, expect to get a good survival rate of cardiac cell transplantation . Chitosan, citric acid and ascorbic acid can be synergistic antioxidant response of the original tissue damage and oxidative stress caused by foreign objects. Chitosan has an excellent biocompatibility, citric acid is a natural body's substances, and ascorbic acid is a necessary body nutrients, three combined togeter to ensure the biocompatibility. Transferring the transplanted cells into the damage tissue by injection can reduce the hazards of surgery pain. The non-flowable semi-solid state hydrogel after injection can support of the transfer cells. With the growth of transplanted cells, scaffold gradually degrades excreted and eventually rebuilds cardiac function.
