中文摘要: 离子交换层析是一种广泛应用于生物大分子的分离、纯化和精制的高效分离方法。蛋白质在离子交换层析介质表面的吸附行为是国内外的研究热点。研究蛋白质在层析介质上的吸附行为,能从分子水平上研究吸附-解吸过程,有助于深入了解离子交换层析的反应机理,从而制备高效新型的离子交换层析介质。本项目用溶菌酶、牛血清白蛋白、免疫球蛋白三种球蛋白作为模型蛋白,研究模型蛋白在DEAE阴离子交换层析介质表面的吸附行为,同时探索形成有利于保持其生物活性的空间结构的调控方法。本项目利用LB膜(Langmuir-Blogett Film)技术建立DEAE琼脂糖膜,通过液质联用和蛋白酶切技术结合研究吸附蛋白质的结合位点,通过石英晶体微天平和原子力显微镜研究吸附蛋白质的空间结构信息,实现对离子交换层析中蛋白质吸附状态的直接检测,研究蛋白质吸附行为的变化规律及调控方法,为离子交换层析纯化过程提供理论指导。

英文摘要: Ion exchange chromatography is widely used in the separation,purification and refinement of biological macromolecules. The protein adsorption behavior on the surface of ion exchange chromatography media has been a hot topic at home and abroad. To reseach the adsorption behavior can study adsorption -desorption proess of ion chromatographic medium on the molecular levels, contribute to a better understanding of the reaction mechanism of ion exchange chromatography and help to prepare the new type of ion exchange chromatography media. This project will use lysozyme, bovine serum albumin and immunoglobulin as model proteins to research the adsorption behavior on the surface of DEAE anion exchange chromatography medium and explore methods to maintain the spatial structure with high biological activity. This project uses the LB films (Langmuir-Blogett Film) technology to establish a DEAE agarose film. Then studies adsorbed protein binding sites by LC-MS and protease cut and protein adsorption space structure by quartz crystal microbalance and atomic force microscopy to in situ detect the protein adsorption behavior on the interface of ion exchange chromatography. All these can serve as guide studies of ion exchange chromatography purification process.
