中文摘要: 降低血液胆固醇水平对防治动脉粥样硬化(AS)和维护人体健康具有重要意义。我们的前期研究证实中链脂肪酸可降低超重者体重、体脂肪,降低血甘油三酯、总胆固醇以及升高血液高密度脂蛋白及高密度脂蛋白胆固醇水平,并进一步获得自然科学基金面上项目(8117266)等支持探讨中链脂肪酸通过促进胆固醇逆转运(RCT)而减少机体胆固醇聚集,发挥抗AS的作用。基于此,本项目假设中链脂肪酸通过调节胆汁酸代谢包括调节肝脏胆汁酸合成、胆固醇及胆汁酸代谢产物排出和胆汁酸肠肝循环,协同RCT途径而降低机体胆固醇的水平。选择雄性C57BL/6J小鼠为研究对象,采用生物化学、色谱分析、ELISA、实时定量PCR、Western blotting等实验技术,观察中链脂肪酸中的辛酸和癸酸对胆汁酸代谢产物的影响以及分析辛酸和癸酸对胆汁酸代谢过程中相关酶、调节蛋白以及核受体表达的影响,探索中链脂肪酸调节胆汁酸代谢而降低胆固醇的作用。

英文摘要: It is essential that reduction of cholesterol is important for control of atherosclerosis(AS) and prevention from cardiovascular disease. Our previous studies have shown that medium-chain fatty acid(MCFA,octanoic acid and decanoic acid) can reduce body weight, blood triglyceride and cholesterol, increase high-density lipoprotein levels in onverweight and hyperglycerdemic subjects, and we have obtained the two grants of the Innovation Fund of the Chinese PLA General Hospital (11KMM44) and the National Natural Science Foundation (81172667) to explore the biological effects of MCFA on atherosclerosis by promoting reverse cholesterol transport (RCT). In this project, we will continue to explore the mechanism of MCFA on regulating bile acid metabolism(synthesis of bile acid in liver,excretion of cholesterol and bile acid , and enterohepatic circulation of the bile acid). The male C57BL/6J mice will be housed to diets containing octanoic acid and decanoic acid. The means of biochemical, chromatographic analysis, ELISA, real-time quantitative PCR, Western blotting techniques will be applied.
