中文摘要: 甲状腺激素是参与人体代谢调控的重要激素之一,主要通过与受体结合发挥作用。临床上甲状腺激素水平异常导致的胰岛素抵抗十分常见,而甲状腺激素促进胰岛素抵抗的分子机制目前仍未完全阐明。本课题组发现,甲状腺激素可以直接作用于肝细胞,刺激Fetuin A的合成和分泌。既往研究证实,Fetuin A可直接与胰岛素受体结合并引起胰岛素抵抗。因此,我们推测 Fetuin A可能在甲状腺激素诱发的胰岛素抵抗中发挥重要作用。本课题组发现:甲状腺激素主要通过TRβ受体发挥对Fetuin A编码基因的转录调控作用;人体内的血清甲状腺激素与Fetuin A水平呈正相关。后续研究中,我们将充分利用分子生物学手段及转基因模式动物,探讨甲状腺激素调控Fetuin A表达的分子机制,并研究fetuin A在甲状腺激素导致的胰岛素抵抗中所扮演的角色,为寻找改善胰岛素抵抗的治疗策略提供思路。

英文摘要: Thyroid hormone is one of the major hormones regulating metabolism, whose function is mediated by its receptors(TR). Insulin resistance is a common phenomenon in patients with disregulated thyroid hormones, of which the molecular basis is still unclear. Recently, our group has discovered that thyroid hormone could directly stimulate hepatocytes to synthesize and secrete Fetuin A, a glycosylated protein previously confirmed to bind insulin receptor and induce insulin resistance. We speculate that Fetuin A might be a mediator of thyroid hormone-induced insulin resistance. Our preliminary data have suggested that the transcription of Fetuin A is stimulated by thyroid hormone mainly via TRβ. Furthermore, the serum Fetuin A is positively correlated with thyroid hormone level. Next, we will try to identify the thyroid hormone response element(TRE) located on the promoter of Fetuin A and delineate to what extent Fetuin A is involved in mediating thyroid hormone-induced insulin resistance. We believe these findings could contribute to the quest for new strategies dealing with insulin resistance.
