中文摘要: 在骨临床上,钛植入体与周边组织整合性差,并且周边组织容易发生缺氧坏死现象。本项目旨在提高钛植入体与骨整合性,同时在钛材表面建立血管网络,为钛周边组织提供氧及营养物质并排除代谢产物。本项目的核心是结合利用接触引导引起的细胞增殖和生长因子诱导的骨髓间充质干细胞(BMSCs)定向分化。通过微弧氧化技术在钛材表面制备具有多孔结构且结合紧密的羟基磷灰石/二氧化钛混合涂层,并通过石蜡覆膜、激光雕刻和氢氟酸腐蚀在涂层上制备血管网络结构;在涂层上引入羟基磷灰石/骨形态发生蛋白(HA/BMP-2)微球和接枝血管内皮生长因子(VEGF)。改性后的钛材在与BMSCs接触时,HA/BMP-2诱导BMSCs向成骨细胞分化,促进新生骨组织形成,提高钛材的骨整合性;而VEGF诱导BMSCs向内皮细胞分化,促进血管网络形成,为新生骨组织提供营养物质和排除代谢产物。项目可为钛基生物材料的表面改性提供新思路。

英文摘要: In clinical, the titanium implant has the poor ability of integration with the surrounding tissue. Furthermore, it will result in the death of the surrounding tissue because of hypoxia. Therefore, in this project, we prepare to improve the ability of integration. Meanwhile, we want to build the blood system on the surface, which can provide oxygen and nourishment for the surrounding tissue and excrete the metabolic wastes. The contact guidance, as well as the growth factors, will be applied to promote the proliferation and the differentiation of bone mesenchymal stem cells. The coating of hydroxyapatite and titanium dioxide will be prepared on the surface of titanium by the method of microarc oxidation. The blood network will be produced on the coating after covered with paraffin, carved by laser and erode by hydrofluoric acid. Furthermore, the microsphere of HA/BMP-2 and VEGF will be grafted on the surface. With these compositions and structures, when the implant is cultured with mesenchymal stem cells, the HA and BMP-2 can induced BMSCs to differentiate into osteoblasts, and promote the formation of new bone. On the other hand, VEGF stimulate BMSCs to differentiate into endothelial cell and create the blood system on the surface of implant, which can provide oxygen and nourishment for the surrounding tissue a
