中文摘要: 本项目针对生理活性分子及离子的特异性检测,结合光致发光和电化学发光分析的优点,拟从高效发光试剂联二吡啶钌(II)配合物入手,依据分子识别的设计理念,通过对联二吡啶配体进行化学修饰,偶联对生理活性组分具有专属识别反应的官能团,设计合成可用于荧光及电化学发光双信号特异性检测的单线态氧、巯基氨基酸、汞离子等生理活性组分的新型发光探针,并研究其光化学、电化学及光致发光与电化学发光性能,实现复杂生物样品中生理活性组分的高灵敏度及高选择性荧光和电化学发光双信号检测。新型联二吡啶钌(II)配合物衍生物发光探针的研究改进了目前荧光探针与电化学发光探针研究与应用中的单模式测定方式,拓展了现有电化学发光试剂的应用范围,不仅可为生物医学与生命科学研究中生理活性组分的测定提供新的分析检测手段,也可为电化学发光分析技术的发展与应用及光-电发光联用技术的发展提供一种有效的策略与理念。

英文摘要: This program focuses on highly specific detection for bioactive species using Ru(II)-bipyridine complexes as probes with the advantages of photoluminescence and electrochemiluminescence (ECL) methods. Based on the molecular recognition theory, some probes for single oxygen, thiol-containing amino acids, or Hg2+ can be designed and synthesized possessing bipyridine ligand modified with specific recognition functional groups towards analytes. The luminescence and ECL properties of these probes will be investigated in order to successfully detect bioactive species in complicated biological samples with high selectivity and sensitivity by dual-signaling luminescence and ECL. The studies on novel probes based on Ru(II)-bipyridine complexes will advance the existing detection model in fluorescent or ECL field from single model to dual, or even multi model, and extend the analytical range of ECL luminophors. This program not only provides some new detection methods for the researches on bioactive species in biomedicine and life sciences, but also offers an effective strategy for detection technology of ECL and its application. It will contribute to the development of combined analytical technology of photo and electric luminescence assay.
