中文摘要: 膀胱癌是我国最常见的泌尿系统肿瘤,而microRNA发挥基因表达调控,参与肿瘤细胞增殖、分化、转移、凋亡、周期调控等生物学过程已得到广泛证实。我们前期研究发现具有抑制肿瘤演进的LASS2基因在不同分期、分级和预后的膀胱癌组织以及不同侵袭潜能的膀胱癌细胞株中存在表达差异,进而运用芯片和生物信息学技术筛选并验证与LASS2基因存在相互作用的最相关的microRNA为hsa-miR-3658。本研究将进一步明确筛选出的hsa-miR-3658对LASS2基因转录后调控翻译表达的影响,对膀胱癌生物学行为的影响;明确hsa-miR-3658对膀胱癌化疗敏感性的影响;发掘与LASS2存在相互作用hsa-miR-3658在预防和治疗膀胱癌中的作用,评估hsa-miR-3658及其靶基因LASS2应用于临床基因治疗的安全性、可行性和有效性,了解其开发的应用价值,为膀胱癌发生发展机制研究提供新的实验依据。

英文摘要: Bladder cancer is one of the most common cancers and microRNAs have been identified as key molecules in regulatory networks, and they involve in tumor cell proliferation, differentiation, metastasis, apoptosis and cycle regulation. The expression of Homo sapiens LAG1 longevity assurance 2 (LASS2) was significantly correlated with tumor stage, cell proliferation, metastasis and invasion. In our previous study, we found that the most relevant LASS2-related microRNAs was hsa-miR-3658 by using the methods of gene chip and bioinformatics technique. The roles of the screened hsa-miR-3658, including post-transcriptional regulation, cell proliferation ability, invasion, metastasis, apoptosis, epithelial-mesenchymal transition and chemosensitivity of bladder cancer cells, will be explicit. This study was designed to study the anticancer effect of hsa-miR-3658 and LASS2 to evaluate its latent energy in prevention and clinical application of human bladder cancer.
