中文摘要: 阿尔茨海默病(Alzheimer's disease,简称AD)造成认知功能丧失的一个重要机制是神经元细胞内钙离子水平调节的失衡。这种钙失衡导致了成熟的神经突触数量减少,突触可塑性减退,神经功能受到严重损害。PTEN是具有强大作用的细胞内钙离子调节器,具有调节细胞内质网钙库和细胞膜离子通道的双重功能。本项目围绕PTEN对神经元突触形成及钙信号的调节这一核心问题,利用转基因AD小鼠模型以及PTEN基因敲除小鼠研究PTEN介导的信号在AD病理过程中的作用。我们将采用体内、体外钙成像技术研究AD病理条件下PTEN对神经细胞内钙离子水平和神经元的树突棘形态学变化等调节的分子机制,明确PTEN及其下游分子对AD中的神经元细胞内钙信号和神经突触结构稳定性的重要作用。本研究将有助于深入理解AD的发病机制和发现治疗AD及老龄记忆衰退的新靶点。

英文摘要: The abnormal intracellular calcium homeostasis is an important mechanism for memory loss in Alzheimer's disease (AD). The calcium abnormality leads to a decrease of mature synapses and synaptic plasticity, which causes damage of neuronal function. PTEN serves as a potent calcium modulator and play dual roles on both ER and cell membrane in modulation of calcium channels. Our proposed studies will focus on PTEN mediated synapse formation and calcium signals and study the role of PTEN mediated signals in AD by using transgenic AD animal model and PTEN knochout mice. We plan to carry out in vivo and in vitro calcium imaging studies to clarify how PTEN modulate calcium signaling and neuronal spine morphogenesis in AD, and to reveal the roles of PTEN and its downstream molecules for calcium signaling and synaptic structure stabilization in AD. Our studies may lead to an increased understanding of new mechanisms and a novel potential therapeutic target for treatment of AD and age-related memory decline.
