中文摘要: 成骨活性差和血管化困难仍是目前骨组织工程研究中的重要问题,3-D细胞培养和载体微型化是解决该问题的有效途径。本项目拟以多孔磷酸钙微球为研究对象,对其作为3-D细胞载体的性能和规律进行基础科学研究。具体内容包括:以矿化后的脱胶桑蚕丝短纤维为模板来制备兼具小孔/大孔连通性的多孔微球载体,以满足营养物质和代谢废物的运输需求,并为细胞的长入、迁移及在载体内的集落生长提供空间和通路;研究模板的添加量、模板矿化过程、成型工艺等因素与所制得微球微结构之间的定量关系;在微球上负载药物/生长因子,通过表面改性方法构建药物/生长因子在微球上的浓度梯度分布,以抑制药物初始突释现象,并延长药物的释放周期;以骨髓间充质干细胞为种子细胞,研究其在微载体上的吸附、迁移、增殖、分化和集落生长行为,并探索微载体的自组装过程及规律。本研究可为骨组织工程用磷酸钙微载体的制备和应用提供科学依据和实验参考。

英文摘要: Poor vascularization and osteogenesis of the implant have been two major problems in the bone tissue engineering. The cultrue of cell in the three-dimensional (3-D) system and the development of microscale technology open new opportunities for improving these two questions.The project focus on the fabrication and properties of porous calcium phosphate-based 3-D microcarrier for cells. The major content includs: (1)Modified degumming silk fiber is used as template to prepare microcarrier with interconnected porous structure. Effect of parameters, including content and mineralization of template, moulding process et al., on microstructure of carrier will be researched respectively; (2)Drug and growth factor for improving differentation of MSCs towards osteoblast-like cell and endothelial cell will be loaded on the surface of calcium phospahte-based microcarrier and amino-ploly(ethylene glycol) will be used to coat the microcarrier in order to controll drug release speed and period;(3)MSCs will be used as cell model, which are seeded on the microcarrier, and then attachment, proliferation, differentation and colony growth of MSCs on the microcarrier will be explored as well as microassambly behavior of microcarrier.
