中文摘要: G-四链体是人端粒末端和基因启动区的富含鸟嘌呤(G)的单链DNA序列形成的四链结构。高特异性G-四链体配体可能具有的高抗肿瘤活性激发了人们对选择性G-四链体配体的探求。杂杯杂芳烃等新型大环化合物以其柔性的构象,独特的空穴结构,可调控的空腔大小等特征很有可能发展成为理想的G-四链体高选择性配体。本项目拟在探索大环分子对G-四链体结构的特异性识别机理的基础上,通过对大环分子的构象调控,实现对特定构型的G-四链体结构的高结合度、高选择性识别,有望应用于G-四链体结构相关的疾病诊疗,同时有望开辟G-四链体结构应用的新领域。

英文摘要: G-quadruplex is a four-stranded structure formed from G-rich overhangs in the human telomere end and the gene promoter regions. The high anti-cancer activity of its specific ligand arouses researchers' interest in the study of highly selective ligands for G-quadruplex. Novel macrocyclic compounds (e.g. heteroatom bridged calix[n]aromatics) have special size of cavity and flexible conformation, which enable them promising highly specific ligand of G-quadruplex. Base on the recognition mechanism study of the macrocycles and G-quadruplexes, this proposal will focus on the recognition of specific G-quadruplexes configurations via specific superior conformations of one macrocycle with conformation regulation. The result will provide new routes for G-quadruplex-related clinics, and open up new application of the G-quadruplex structure.
