中文摘要: 药用植物的有效成分是在一系列关键酶的催化下生成的,这些酶并非孤立的存在于细胞中,而是与其它蛋白通过相互作用的方式行使其功能。丹参牻牛儿基牻牛儿基焦磷酸合酶(SmGGPS)是控制丹参酮类化合物生物合成途径中的一个关键分支点,为揭示(SmGGPS)在丹参酮类化合物生物合成过程中的作用机理,本项目从蛋白质相互作用的角度出发,以SmGGPS为"诱饵"蛋白,通过酵母双杂交系统筛选丹参cDNA文库,得到了与SmGGPS存在相互作用的候选蛋白,为进一步明确与SmGGPS存在相互作用的蛋白,将通过双分子荧光互补技术和细胞内共定位的方法进行确证蛋白间的相互作用,并对互作蛋白和SmGGPS进行RNAi和过表达分析,揭示互作蛋白对SmGGPS功能的影响,进一步解析蛋白间的互作关系对丹参酮类化合物代谢的调控机理。

英文摘要: A series of enzymes involved in the bioactive components of medicinal Plants biosynthesis,existed not alone in cells and interacted with other proteins to exercise functions. GGPS1 has been considered as an important regulatory target in the tanshinone biosynthetic pathway. To illustrate the mechanism of SmGGPS involved in tanshinone biosynthesis, the proteins interacted with SmGGPS were screened the cDNA library by yeast two hybrid technology and verified by BIFC and colocali zation in cells. To further reveal the metabolism of the relationship between the interacted protein and SmGGPS on tanshinone compounds biosynthesis,the interacted protein and SmGGPS were transferred into Salvia miltiorrhiza h by RNAi and overexpression to explore the interacted protein affected on the function of SmGGPS.
