中文摘要: 长链非编码RNA(lncRNA)在哺乳动物的神经系统中广泛表达,对神经系统的发育具有极其重要的作用。目前人们采用全脑或不同神经解剖学区域来研究神经系统lncRNA。由于同一神经解剖学区域细胞的不均一性,上述研究不能检测到在神经系统特定细胞类群低丰度表达且起关键调控作用的lncRNA。我们通过单细胞高通量深度测序,首次从小鼠侧脑室室管膜下区(SVZ)鉴定了600多个新的细胞类型特异的多外显子lncRNA,并采用WGCNA方法找出了9个与干细胞干性维持和分化相关的蛋白质有关联的lncRNA。本项目拟用原位杂交技术和定量PCR方法比较这些lncRNA在小鼠脑内和体外培养的细胞中的表达谱,通过过表达或表达下调、以及制备lncRNA基因敲除鼠在体外和体内研究这些lncRNA的功能,揭示lncRNA在干细胞干性维持和分化中的作用机理,构建lncRNA与蛋白质和转录因子相互作用的调控网络。

英文摘要: Long noncoding RNA (lncRNAs) are expressed widespread in mammalian central nervous system and have key regulatory and functional roles in nervous development. At present, lncRNAs in nervous system are investigated in whole brain or different neuroanatomical regions, which can't identify low abundance lncRNAs expressed in specific cell types. By single-cell whole transcriptome analysis, we identify more than 600 novel multi-exonic lncRNAs from postnatal forebrain subventricular zone in mice and found 9 lncRNAs associated with protein coding genes involved in the maintenance of stemness and differentiation in adult neural stem cells. To investigate the function of these lncRNA, we will compare lncRNA expression profile in different neuroanatimical regions and cultured cells by fluorescence in situ hybridization and real-time PCR and study lncRNA's function through overexpress, knockdown and lncRNA gene knockout mice. Understanding pronounced trends in lncRNA expression and their potential proteins or trascription factors interactions will provide valuable insight into the molecular mechanisms of stemness maintenance and differentiation in neural stem cells.
