中文摘要: 为了明确1,25(OH)2D3在抗皮肤和骨骼衰老中的作用及机制,我们拟利用抗氧化功能严重障碍的Bmi-1基因敲除纯合子小鼠作为研究模型,分别给予1,25(OH)2D3和抗氧化剂,利用组织病理学、细胞生物学和分子生物学的方法,比较分析各实验组动物皮肤和骨骼的表型变化,包括细胞增殖、凋亡、衰老和新生血管发生以及成骨细胞骨形成和破骨细胞骨吸收相关指标的检测,观察1,25(OH)2D3是否与抗氧化剂一样,能够部分或完全纠正Bmi-1基因缺失纯合子小鼠的皮肤和骨骼衰老表型;利用流式细胞仪技术、蛋白质印迹、实时荧光定量RT-PCR以及免疫组化等方法检测上述动物的皮肤和骨骼的氧化应激以及DNA损伤相关指标,观察1,25(OH)2D3是否能够通过提高抗氧化作用,延缓皮肤老化和骨骼衰老。本研究不仅有助于阐明活性维生素D在抗衰老中的作用机制,而且将为活性维生素D运用到延缓衰老的临床研究提供理论和实验依据。

英文摘要: To determine whether 1,25(OH)2D3 plays a role in preventing aging in skin and skeleton,the homozygous Bmi-1 gene knockout mice with severe defects in antioxidative function used as aging model will be supplemented with 1,25(OH)2D3 or antioxidant.Phenotypes of skin and skeleton from mice will be analyzed including cellular proliferation, apoptosis and senescence and angiogenesis,osteoblastic bone formation and osteoclastic bone resorption related parameters,by techniques of histopathology, cellular and molecular biology to assess whether 1,25(OH)2D3 and antioxidant can rescue the premature senescence phenotypes in skin and skeleton of homozygous Bmi-1 gene knockout mice. The oxidative stress and DNA damage related parameters will be examined in skin and skeleton by flow cytometry, Western blots, Real-time RT-PCR and immunohistochemistry to determine whether 1,25(OH)2D3 can play a role in postponing skin and skeletal aging through its antioxitative action. Results from this study will be helpful to elucidate mechanisms of active vitamin D in preventing aging and will provide theoretical and experimental basis for the applications of sufficient vitamin D in clinical study on postponing aging.
