中文摘要: 植物细胞内的化学组成信息对于研究植物细胞代谢、分化、疾病以及生态效应等具有重要的指示作用。不同细胞受年龄、营养、细胞循环周期以及环境因素的影响,体内的新陈代谢产物存在很大的差异。植物细胞体积小、细胞内成分复杂,直接分析活体植物细胞中的生物分子对于分析工作者来说是一个很大的挑战。本课题提出了一种基于活体纳喷雾质谱的新方法,用于活体植物单细胞中生物分子的分析。拟采用一根充满液体的毛细管插入细胞内充当液体电极,另一根毛细管进行植物细胞的原位取样以及在线离子化,细胞内目标分子被离子化后进入质谱进行检测。选取洋葱表皮细胞为研究对象,利用该方法分析洋葱表皮细胞中的氨基酸类,确定氨基酸在不同细胞中的分布情况。本项目的完成有望为活体植物单细胞分析提供一种新思路和新手段,将在植物生理生态研究中具有潜在的应用前景。

英文摘要: Information on the chemical composition within a cell has implications in the understanding of cell metabolism, division, disease states, ecological effects, etc. Depending on age, phase in the cell cycle, nutrition, and environmental factors, individual cells exhibit large metabolic diversity. It is a big challenge for analyst to detect the biomolecules in single plant cell, because it has a small volume but complex compounds. Here we proposed a new method for the analysis of biomolecules in single plant cell based on the in vivo nanoESI mass spectrometry. A capillary fill with water is inserted into the plant cell and acts as a liquid electrode. The second capillary is used for sampling from the cell in situ, and then the sample is ionized on line. The target ions are transformed into the mass spectrometry for detection. We plan to identify the amino acids in onion epidermal cell and obtain their distribution in different plant cells with this method. This item will provide a new idea for the analysis of single live plant cell, which has potential applications in the studying of plant physiology and ecology.
