中文摘要: 非结核分枝杆菌(NTMs)与结核杆菌同属分枝杆菌属,其单独或与结核杆菌共感染均引起人和动物发病,且发病率不断上升,危害越来越严重。目前,有关NTMs感染的分子免疫机制尚不清晰。本研究在已分离到的不同动物源NTMs和确证其影响牛结核免疫效果的基础上,拟建立NTMs感染C57BL/6小鼠和人源化小鼠动物模型;应用抗体中和、流式细胞术、酶联免疫法等免疫学技术研究屏蔽调节性T细胞(Tregs)对NTMs感染小鼠慢性感染的影响;缺乏CTLA-4、IL-10和TGF-β时,小鼠感染NTMs后细胞因子的表达、病理变化以及细菌量等指标的变化,据此分析CTLA-4、IL-10、TGF-β等对Tregs在慢性感染免疫调节中的作用,旨在初步揭示Tregs参与NTMs慢性感染分子免疫调节机制,为进一步丰富NTMs基础免疫学理论、提高疫苗免疫效力具有重要意义。

英文摘要: Nontuberculosis mycobacteria and Mycobacterium tuberculosis both belong to mycobacterium,its solo infection or coinfection with Mycobacterium tuberculosis can cause morbidity of human beings and animals,moreover,the morbidity rate upgrades gradually and the hazard become more and more serious.Now,the molecular immune mechanism of Nontuberculosis mycobacteria infection is not clear.On the basis of Nontuberculosis mycobacteria separated from different animals and authenticating its influence on bovine tuberculosis immunoeffect,this research will establish animal models of C57BL/6 and humanize mouse. In order to reveal the molecular immune regulation mechanism of Tregs in Nontuberculosis mycobacteria chronic infection initially, first of all, the influence of screening Tregs on Nontuberculosis mycobacteria chronic infection will be studied with antibody neutralization, flow cytometry, ELISA, etc. Moreover, the expression of cytokine, pathological change and bacterium quantity after Nontuberculosis mycobacteria infection will be analyzed in lack of CTLA-4,IL-10 and TGF-β. I believe that the mechanism of Tregs molecular immunoregulation during Nontuberculosis mycobacteria chronic infection will be revealed in this study.
