中文摘要: 在前期制备切割穹窿海马伞大鼠模型、海马基因芯片筛选的基础上,从海马神经再生角度,选择了与诱导神经干细胞迁移有关的基质细胞衍生因子-1α(SDF-1α)、诱导神经干细胞向成熟神经元分化的胰岛素样生长因子-Ⅱ(IGF-Ⅱ)及促使放射状胶质细胞增殖和向神经元分化的矮小相关转录因子1(RUNX1T1),在切割穹窿海马伞大鼠模型及用切割侧海马提取液在细胞培养中模拟海马神经再生微环境,探讨海马放射状胶质细胞SDF-1α、IGF-Ⅱ及RUNX1T1 mRNA和蛋白的表达变化,以及与海马神经干细胞增殖、迁移、向神经元分化和放射状胶质细胞增殖和向神经元分化的关系;并在体内外的海马放射状胶质细胞中沉默或过表达上述3个基因,观察对诱导神经干细胞增殖、迁移和向神经元分化及放射状胶质细胞增殖和向神经元分化的影响,以明确放射状胶质细胞在海马神经再生过程中所起的作用及其作用机制,为临床治疗认知功能障碍疾病提供新思路。

英文摘要: This study is on the basis of previous preparation of fimbria-fornix transected rats model, hippocampal gene chip screening and preliminary experiments, from the view of hippocampal neural regeneration, three factors will be selected: stromal cell derived factor -1α(SDF-1α) which is related to the neural stem cells migration, insulin like growth factors-Ⅱ(IGF-Ⅱ) which induce neural stem cells differention into mature neurons and runt related transcription factor 1 (RUNX1T1) which promote radial glial cells proliferation and differention into neurons. Using fimbria-fornix transected rats model and the extracts from fimbria-fornix transected hippocampus to mimic the hippocampal neural regeneration niche, to investigate the dynamic expression of SDF-1α, IGF-Ⅱ, RUNX1T1 mRNA and protein in the radial glial cells, and its relationship to hippocampal neural stem cells proliferation, migration and neuronal differentiation including the radial glial cells neuronal proliferation and differentiation; then silence or over-express above mentioned three genes in hippocampal radial glial cells both in vivo and in vitro, to investigate their effects on the proliferation and migration of neural stem cells and neuronal differentiation including the proliferation and neuronal differentiation of radial glial cells, to identify the
