中文摘要: 重金属汞是一种常见的环境污染物,诱导以T细胞依赖的多克隆B细胞活化为特征的自身性免疫反应,但是其发病机理仍然不清楚。树突状细胞是专职的抗原提呈细胞,又是一种免疫调节细胞,对抑制自身免疫反应起重要的作用。目前有关汞对体内树突状细胞影响的研究尚未见报道。因此,我们将利用三种对汞免疫毒性敏感度不同的小鼠,即SJL(高度敏感)、BALB/c(敏感)、DBA/2(不敏感),系统地研究汞对树突状细胞及对其亚群构成的影响,以及汞对树突状细胞的免疫调节分子及其转移能力的影响。另外,我们将以呼吸道接触卵清蛋白(OVA)诱发过敏性哮喘为模型,研究汞对机体过敏性反应的影响以及树突状细胞在此反应中的作用。本课题的研究,对深入了解汞的毒性及毒作用机制,对预防和治疗汞对健康的影响提供新的科学依据。

英文摘要: Heavy metal mercury is one of widely spread environmental pollutants. Mercury induces autoimmunity characterized by a T cell-dependent polyclonal activation of B cells. However, its mechanism remains unclear. Dendritic cells (DCs) are professional antigen-presenting cells, and are immuno-regulatory cells that play an important role in modulating autoimmunity. So far, there is no report describing the effects of mercury on the behaviors of DCs in vivo. Using three different stain mice that have different susceptibility to mercury-mediated immunotoxicity, i.e., SJL (highly susceptible), BALB/c (susceptible), DBA/2 (resistant), we will systemically investigate how mercury modulates DCs in vivo, including the proportion of DCs, up- and down- regulation markers of DCs, and DC's ability of migration to draining lymph nodes. Furthermore, we will determine whether exposure of mercury enhances the allergic response in OVA-induced asthma model mice, and the possible roles of DCs in this reaction. Our study will help us understand more details about mercuric immunotoxicity and its mechanisms, and will provide new suggestions and evidences for preventing and treating mercury-related diseases.
