中文摘要: 羊绒是皮肤次级毛囊产生的纤细底层毛,具有重要的经济价值。羊绒的生长是一个复杂的过程,具有明显的周期性变化,受多个信号通路和调控因子的影响,而且非编码RNA参与转录后的调控过程。长链非编码RNAs(long noncoding RNAs,lncRNAs)是一类本身不编码蛋白、转录本长度超过200nt的RNA分子,可以在表观遗传调控、转录调控及转录后调控等层面上调控基因的表达。哺乳动物基因组中许多lncRNAs仅在特定发育阶段出现, 或具有组织细胞特异性。目前,关于lncRNAs调控羊绒生长的研究尚未见报道。本项目以陕北白绒山羊为研究对象,拟通过lncRNA芯片及生物信息学方法,筛选羊绒生长期和休止期差异表达的特定lncRNAs,预测和验证其作用靶点;利用过表达和siRNA干扰验证特定lncRNA的功能,从lncRNAs调控的角度阐述羊绒周期性生长的分子机理,为绒山羊进一步的育种工作奠定基础。

英文摘要: Cashmere is the fine undercoat produced by the secondary follicle in the skin of cashmere goat. It has important economic value in husbandry industry. The growth of cashmere is a complex process and has significant periodic changes affected by many signal pathways and regulators. Non-coding RNA also involves in the post transcriptional regulation of hair growth. lncRNAs (Long non-coding RNAs) are in general considered as non-protein coding transcripts longer than 200 nucleotides. They can regulate gene expression in epigenetic regulation, basal transcription and post-transcriptional regulation. Many lncRNAs in mammalian genome sequence appear in specific developmental process or have tissue and cell type specific. There have no report on lncRNA regulating cashmere growth until now. The proposal will screen the differentially expressed specific lncRNAs in anagen and telogen of Shanbei cashmere goat by using lncRNA array and bioinformatics methods. The target binding site of specific lncRNAs will be predicted and identified, and their functions will be verified by over-expressing and RNA small interference. The periodic growth mechanism of cashmere will be elaborated from the aspects of lncRNA regulation, which will provide reference for the further breeding work of cashmere goat.
